چکیده

The aim of this study was to investigate the protective role of red grape seed extracts (GSEs) against oxidative damage on DNA, brain and erythrocytes in vitro. In this study, the protective effects of two GSEs (water and ethanol) on free radical mediated DNA-sugar damage, Fe -induced lipid peroxidation in brain and 2+ H O induced hemolysis and lipid peroxidation of erythrocytes were determined. The results revealed that the 2 2 presence of various concentrations of each grape seed extract in the reaction mixture prevented the free radical-mediated DNA-sugar damage in a dose dependently. Similarly, GSEs caused a significant decrease in Fe -induced lipid peroxidation in rat brain tissues in a dose dependent manner. Both GSEs at different 2+ concentrations suppressed H O -induced hemolysis of rat erythrocytes in a concentration-dependent manner. 2 2 Pre-treatment of cells with GSEs, however, prevented the phospholipid oxidative alteration in a concentration-dependent manner. Noteworthy, the ethanol grape seed extract (EGSE) was more effective than water grape seed extract (WGSE) against oxidative damage of DNA, brain and erythrocytes. In conclusion, the results demonstrated that GSEs protected DNA, brain and erythrocytes against oxidative stress and it could be used as a valuable food supplement or a nutraceutical product.

Neuroprotection of Grape Seed Extract and Pyridoxine against Triton-Induced Neurotoxicity

Triton WR-1339 administration causes neurotoxicity. Natural products and herbal extracts can attenuate cerebral injury. In the present study, we investigated the neuroprotective role of grape seed extract and/or vitamin B6 against triton-induced neurotoxicity. Thirty-five adult male albino rats of the Sprague-Dawley strain, weighing 140-145 g, were divided into five groups: control, triton, gra...

Scavenging and antioxidant properties of different grape cultivars against ionizing radiation-induced liver damage ex vivo.

Ionizing radiation (IR) has become an integral part of the modern medicine--both for diagnosis as well as therapy. However, normal tissues or even distant cells also suffer IR-induced free radical insult. It may be more damaging in longer term than direct radiation exposure. Antioxidants provide protection against IR-induced damage. Grapes are the richest source of antioxidants. Here, we assess...

Protective Effect of Quercetin on Oxidative Stress in Glucose-6-Phosphate Dehydrogenase-Deficient Erythrocytes in Vitro

Glucose-6-phosphate dehydrogenase (G6PD) deficient subjects are vulnerable to oxidative stress. Quercetin, a flavonoids, has been employed as a potent oxygen-free radical scavenger in order to assess the protective effects of quercetin against H2O2-induced oxidative damage in G6PD-deficient and normal human erythrocytes. Erythrocytes of G6PD-deficient (n = 10) and normal (n = 10) subjects were ...

Grape Seed Proanthocyanidin Extract Improved some of Biochemical Parameters and Antioxidant Disturbances of Red Blood Cells in Diabetic Rats

Grape seed proanthocyanidin extract (GSPE) has a broad spectrum of biologic properties against oxidative stress. This study aimed to investigate the effects of GSPE on biochemical factors and antioxidant enzymes of erythrocyte in diabetic rats. Diabetes was induced through single injection of streptozotocin (50 mg.kg-1, i.p). Forty Male Sprague-Dawley rats were divided into four Groups: Group 1...
